Rome and Florence are 1.5 hours apart by fast train and most itineraries include both, so the real question is how many nights each deserves and which suits a short trip if you can only pick one.
What Rome gives you
Three thousand years of layered history in one place - the Colosseum and Forum, the Pantheon, the Vatican, Trastevere, and a big, loud, living city around it. It needs more time and energy: distances are longer, crowds are heavier, and the must-see list is deep. Three full days is a sensible minimum.
What Florence gives you
The densest concentration of Renaissance art and architecture anywhere - the Uffizi, the Accademia (David), the Duomo, the Bargello - in a center you can cross on foot in 20 minutes. It's calmer and more manageable than Rome, and it's the gateway to Tuscany. Two days covers the highlights; a third lets you breathe or day-trip.
If you can only visit one
For a first trip to Italy, choose Rome - it's the more complete, once-in-a-lifetime city, and it has range beyond art. Choose Florence if you're specifically drawn to Renaissance art, want a lower-stress city break, or plan to pair it with the Tuscan countryside.
Doing both
Rome (3 nights) then Florence (2 nights), or the reverse, with the Frecciarossa or Italo train between. Book the Vatican, Colosseum, Uffizi, and Accademia with timed tickets in advance for either order - they all sell out in season.
